Abstract

Neisseria gonorrhoeae dan Staphylococcus aureus merupakan dua jenis bakteri penyebab penyakit infeksi. Penelitian mengenai antimikroba dari bahan alami untuk terapi penyakit infeksi mendapat perhatian lebih besar karena efek samping yang sedikit dan potensi terapi yang besar, salah satunya adalah spesies dari Pleurotus. Penelitian ini bertujuan untuk mengetahui profil protein bakteri N.gonorrhoeae dan S.aureus sebelum dan sesudah pemberian ekstrak P.flabellatus. Penelitian ini menggunakan ekstrak P.flabellatus sebagai antibakteri dengan berbagai macam konsentrasi. Bakteri N.gonorrhoeae dan S.aureus sebagai subyek uji. Pemisahan profil protein menggunakan metode SDS PAGE 12%. Hasilnya, bakteri N.gonorrhoeae yang diberi perlakuan ekstrak P.flabellatus memiliki 24-25 pita dengan BM 142 kDa-21 kDa. Bakteri S. aureus yang diberi perlakuan ekstrak P.flabellatus memiliki 21-22 pita dengan BM 146 kDa- 17 kDa. Profil protein bakteri N.gonorrhoeae dan S.aureus yang diberi perlakuan ekstrak P.flabellatus dilihat dari topologinya mirip. Kata kunci: Ekstrak Pleurotus flabellatus, Neisseria gonorrhoeae, Staphylococcus aureus, profil protein Neisseria gonorrhoeae and Staphylococcus aureus are two types of bacteria that cause infectious disease. Research on antimicrobial from natural source for infectious disease treatment got more attention because it has less side effect and great therapeutic potential, for example is species of Pleurotus. This research aimed to know the protein profile of N.gonorrhoeae and S.aureus bacteria before and after treated with P.flabellatus extract. This research is using P.flabellatus extract as antibacteria with any concentration. N.gonorrhoeae and S.aureus bacteria as test subject. Separation of protein profile using SDS PAGE 12% method. The result, N.gonorrhoeae bacteria treated with P.flabellatus extract have protein band varies between 24-25 bands with molecular weight between 142 kDa-21 kDa. S. aureus bacteria treated with P.flabellatus extract have protein band varies between 21-22 band with molecular weigth between 146 kDa- 17 kDa. Protein profile of N.gonorrhoeae and S.aureus bacteria treated with P.flabellatus extract topologically is similar.
