Abstract

Pilli sebagai faktor perlekatan pada mukosa instental, yang sangat penting sebagai awal mekanisme patogenesis dalam proses invasi bakteri menembus dinding sel epitel. Tujuan penelitian mendiskripsikan profil protein pilli E.coli isolat darah pasien widal positif menggunakan metode SDS-PAGE. Peneltian ini dilakukan dengan 3 tahapan yaitu: kultivasi bakteri dengan media bifasik (BHI Agar dan BHI cair), isolasi protein pilli kemudian diseparasidengan SDS-PAGE 10%. Hasil penelitian menunjukkan adanya 28 pita protein, terdapat 9 pita protein yang tampak tebal (pita mayor) dengan berat molekul 100 kDa; 93 kDa; 45 kDa; 44 kDa; 36 kDa; 35 kDa; 21 kDa; 16 kDa; 15 kDa; serta terdapat 19 pita protein yang tampak tipis (protein minor) dengan berat molekul 245 kDa; 209 kDa; 140 kDa; 52 kDa; 51 kDa; 49 kDa; 43 kDa; 41 kDa; 39 kDa; 33 kDa; 32 kDa; 30 kDa; 28 kDa; 27 kDa; 26 kDa; 25 kDa; 23 kDa; 22 kDa; 19 kDa. Kata kunci : Escherichia coli, Protein pilli dan SDS-PAGE, Widal. Pili as an attachment factor of bacteria to intestinal mucus is the first important mechanism of pathogenesis in bacterial invasion through the intestinal cell wall. This research aims to describe characterization of Escherichia Coli’s Pili protein in Blood Isolate of Positive Widal Test Patient by SDS-PAGE method. This research has three steps, the first step is cultivating bacteria with biphasic medium ( BHI agar and BHI liquid), next step is isolating pili protein then separating bySDS-PAGE 10%. The result shows there are 28 protein bands, 9 protein bands are bold ( major band) its molecular weigh are 100kDa; 93 kDa; 45 kDa; 44 kDa; 36 kDa; 35 kDa; 21 kDa; 16 kDa; 15 kDa; 19 protein bands are thin ( minor protein ) ) its molecular weigh are 245 kDa; 209 kDa; 140 kDa; 52 kDa; 51 kDa; 49 kDa; 43 kDa; 41 kDa; 39 kDa; 33 kDa; 32 kDa; 30 kDa; 28 kDa; 27 kDa; 26 kDa; 25 kDa; 23 kDa; 22 kDa; 19 kDa.
