Abstract
Trigliserida adalah lemak utama dalam makanan berperan dalam transport dan penyimpanan lemak dalam tubuh. Kadar trigliserida dalam darah dapat diukur menggunakan alat spektrofotometer maupun POCT (Point of Care Test). Prinsip kerja spektrofotometer dengan mengukur intensitas cahaya berdasarkan absorbansi pada panjang gelombang tertentu dan metode pemeriksaan trigliserida berupa Enzimatik kolorimetri (GPO-PAP).POCT mengukur berdasarkan reflektan (pemantulancahaya) namun, penggunaannya dipengaruhi beberapa faktor seperti suhu dan getaran dan kurang stabil dibandingkan spektrofotometer. Bahan pemeriksaan yang digunakan POCT adalah whole blood dalam jumlah sedikit sehingga sulit diketahui kualitas sampel. Spektrofotometer dapat dilakukan kontrol sehingga mutu (kualitas) hasil dapat diketahui.Tujuan penelitian untuk mengetahui perbedaan kadar trigliserida menggunakan metode POCT dan spektrofotometer.Jenis penelitian adalah analitik cross sectinal. Sampel penelitian berupa 16 karyawanPuskesmas Mranggen 1 Demak. Hasil kadar trigliserida pada alat POCT memiliki rata- rata 132 mg/dl sedangkan dengan spektrofotometer 141 mg/dl. Berdasarkan hasil uji t independent, terdapat perbedaan kadar trigliserida antara alat POCT dan spektrofotometer dengan nilai signifikasi 0,000 < 0,05.Metode pemeriksaan pada alat spektrofotometer adalah Enzimatik kolorimetri sedangkan pada alat POCT adalah reflectance. Kata Kunci : Kadar trigliserida, POCT dan spektrofotometer. Triglycerides are the main fats in the diet have a function in the transport and storage of fat in the body. Levels of triglycerides in blood can be measured using a spectrophotometer or POCT (Point of Care Test). The principle of spectrophotometer by measuring the intensity of light based on absorbance at certain wavelength and triglyceride examination method of Enzymaticcolorimetry (GPO-PAP).POCT measures by reflectant (light reflection) however can beinfluenced by several factors such as temperature and vibration and less stable than spectrophotometer. The examination material used by POCT is whole blood in small amount so it is difficult to know the quality of the sample. Spectrophotometer can be controlled so that the quality (quality) results can be known. The aim of this research is to know the difference of triglyceride level using POCT method and spectrophotometer. The type of research is cross sectinal analytics. The samples were 16 employees of PuskesmasMranggen 1 Demak. The result of triglyceride level on POCT device had mean 132 mg / dl while with spectrophotometer 141 mg / dl. Based on the result of independent t test, there is difference of triglyceride level beetwen POCT and spectrophotometer with signification value 0,000 < 0,05.inspection methoh on spectrophotometer is enzymatic colorimetry while in POCT tool is reflectance.
