Abstract

Salmonella typhi adalah bakteri anggota familia Enterobacteriaceae yang merupakan bakteri enterik dan bersifat batang gram negatif, bakteri ini memiliki alat perlekatan pada sel inang yang disebut pilli, S. typhi merupakan bakteri penyebab demam tifoid yaitu penyakit infeksi menular yang dapat dideteksi menggunakan uji widal, untuk mendapatkan antigen dari bakteri S. typhi dapat dilakukan karakterisasi protein pilli sebagai dasar pengembangan reagen diagnostik yang lebih spesifik untuk uji widal. Tujuan Penelitian ini adalah untuk mengkarakterisasi protein pilli S. typhi SLT-1 serovar H1-j pada kultur darah widal positif dengan SDS-PAGE. Objek penelitian adalah bakteri S. typhi SLT 1 H1-j yang diperoleh dari kultur darah pasien dengan uji widal positif dari Kota Salatiga, kemudian dikultivasi pada media bifasik (BHI agar ditambah BHI cair), isolasi protein pilli metode Ehara, selanjutnya diseparasi dengan SDS-PAGE. Hasil penelitian menunjukkan adanya 1 sub unit protein mayor dengan berat molekul 37 kDa dan 28 protein minor dengan berat molekul 180 kDa; 163 kDa; 155 kDa; 140 kDa; 135 kDa; 124 kDa; 117 kDa; 100 kDa; 75 kDa; 74 kDa; 66 kDa; 65 kDa; 56 kDa; 51 kDa; 45 kDa; 44 kDa; 42 kDa; 40 kDa; 38 kDa; 35 kDa; 33 kDa; 32 kDa; 28 kDa; 27 kDa; 24 kDa; 23 kDa; 22 kDa; 20 kDa. Kata Kunci : Protein Pilli, Salmonella typhi, SDS-PAGE dan Widal. Salmonella typhi is a bacterium belonging to the Enterobacteriaceae’s family which is an enteric bacterium and has a negative stem gram quality, this bacterium has an attachment device on a host cell is called pilli, S. typhi is a bacterium that causes typhoid fever is a contagious infectious disease that can be detected using the widal test, to obtain the antigen of S. typhi bacterium can be characterized pilli proteins as the basic for developing more specific diagnostic reagents for the widal test. The objective of this study was for characterizing the pilli proteins of S. typhi SLT-1 serovar H1-j on a positive widal blood cultures with SDS-PAGE. The research object was S. typhi SLT 1 H1-j bacterium obtained from blood culture of patients with positive widal test from Salatiga City, then it’s cultivated on biphasic media (BHI for added BHI liquid), pilli proteins isolation of Ehara method, furthermore it’s separated with SDS- PAGE. The results showed there was 1 sub unit of major protein with molecular weight of 37 kDa and 28 minor protein with molecular weight of 180 kDa; 163 kDa; 155 kDa; 140 kDa; 135 kDa; 124 kDa; 117 kDa; 100 kDa; 75 kDa; 74 kDa; 66 kDa; 65 kDa; 56 kDa; 51 kDa; 45 kDa; 44 kDa; 42 kDa; 40 kDa; 38 kDa; 35 kDa; 33 kDa; 32 kDa; 28 kDa; 27 kDa; 24 kDa; 23 kDa; 22 kDa; 20 kDa.
