Abstract

HER2 (Human Epidermal Growth Factor Receptor 2) is a receptor on the cell surface that affects on the tissue proliferation. Over expression caused by HER2 were related to cancer. Blocking proteins in IHC staining play a role in minimizing non-specific proteins that compete to bind to existing antibodies in tissues using normal serum and protein (gelatin) solutions. The purpose of this research is to know and analyze the intensity of HER2 by using gelatin concentration of 1.0%, 1.5%, 2.0% and 2.5% as protein blocking also know the best gelatin concentration. Experimental research with cross sectional design. A sample of the research used HER2 3 (+3) positive breast tissue from one patient. IHC-staining used Strep (Avidin) Biotin Complex (ABC) technique using blocking protein (gelatin) concentrate 1,0%, 1,5%, 2,0% and 2,5% with normal serum as a control. The result of HER2 staining using gelatin 1% and 1.5% was obtained the result of positive intensity 2 (+2), 2.0% gelatin obtained the result positive 3 (+3), 2.5% gelatin obtained the result positive 1 (+1) and Normal serum obtained the result positve 3 (+3). There was a significant difference between normal serum with 1%, 1,5% and 2,5% gelatin. There wasn't a significant difference between normal serum with 2% gelatin. The conclusion is the best concentration of 2% gelatin. Keywords: HER2, Blocking Protein, Gelatin
