Abstract
Breast milk is one of the foods that are rich in nutrients. Protein content, glucose, fat, mineral salts cause microorganisms easily grow in breast milk. Lactic acid bacteria is a group of bacteria capable of fermenting lactose into lactic acid. The bactericidal effect of lactic acid is associated with a decrease in the pH of breast milk. The aim of this study was to find out the total lactic acid bacteria in breast milk stored in freezer for 2, 4, and 6 days and to analyze the difference of total lactic acid bacteria in breast milk stored in freezer for 2, 4, and 6 days. The research was conducted descriptively using mother's milk sample as much as 6 lactation mothers. The stages of the study were begun with a multilevel dilution using a 0.85% NaCl diluent solution. Samples were diluted to 10-7 then inoculated on De Man, Rogosa, Sharpel agar medium added to 1% CaCO3 by spread plate method and incubated at 37 ° C for 24 hours. The results showed no growth of lactic acid bacteria. Keywords: Breast milk, Lactic Acid Bacteria Air susu ibu (ASI) merupakan salah satu bahan pangan yang kaya akan zat gizi. Kandungan protein, glukosa, lemak, garam mineral menyebabkan mikroorganisme mudah tumbuh dalam ASI. Bakteri asam laktat (BAL) adalah kelompok bakteri yang mampu memfermentasikan laktosa menjadi asam laktat. Efek bakterisidal dari asam laktat berkaitan dengan penurunan pH ASI. Penelitian ini bertujuan mengetahui total BAL pada air susu ibu yang disimpan pada freezer selama 2, 4, dan 6 hari dan menganalisis perbedaan total BAL pada air susu ibu yang disimpan pada freezer selama 2, 4, dan 6 hari. Penelitian dilakukan secara deskriptif menggunakan sampel ASI sebanyak 6 ibu laktasi. Tahapan penelitian diawali dengan pengenceran bertingkat menggunakan larutan pengencer NaCl 0,85 %. Sampel diencerkan hingga 10-7 kemudian diinokulasikan pada media De Man, Rogosa, Sharpel (MRS) Agar yang ditambahkan dengan CaCO3 1 % dengan metode spread plate dan diinkubasi pada suhu 37oC selama 24 jam. Hasil penelitian menunjukkan tidak terjadi pertumbuhan BAL. Kata kunci : Air Susu Ibu, Bakteri Asam Laktat
