Abstract

Pemeriksaan laboratorium untuk penegakan leukemia salah satunya dengan pewarnaan sitokimia, diantaranya menggunakan reagen lepehne yang dimana reagen lepehne akan tampak warna hijau pada granula eritrosit tua sebagai pembandingnya, lepehne mewarnai sel jajaran eritrosit (eritroblas).Penelitian ini bertujuan untuk mengetahui pengaruh variasi konsentrasi perhidrol (H2O2) terhadap kualitas hasil pengecatan lepehne. Jenis penelitian ini adalah kuantitatif true eksperimen (eksperimen murni). Sampel dalam penelitian ini adalah hasil biopsi sumsum tulang dengan metode sampling random sampling. Berdasarkan hasil pengamatan mikroskopis terhadap lima sampel ditemukan bahwa konsentrasi perhidrol 30% sebagai kontrol terlihat warna hijau jelas pekat, 27,5%(v/v) dan 25%(v/v) menunjukan hasil yang sama yaitu memiliki kualitas pengecatan lepehne yang baik, warna eritrosit hijau tidak pekat. Hal ini menunjukan bahwa penurunan konsentrasi perhidrol 27,5%(v/v) dan 25%(v/v) tidak mempengaruhi hasil kualitas pewarnaan. Sedangkan dengan variasi konsentrasi 22,5% dan 20% menunjukan hasil yang sama yaitu memiliki kualitas yang kurang baik, warna eritrosit hijau tidak jelas. Hal ini menunjukan bahwa penurutan konsentrasi perhidrol yang lebih banyak mempengaruhi hasil kualitas pengecatan lepehne. Berdasarkan analisa data menggunakan Chi-Square, nilai p-value 0,000 < α (0,05) maka disimpulkan bahwa ada pengaruh variasi konsentrasi perhidrol terhadap kualitas hasil pewarnaan lephene. Kata Kunci : konsentrasi perhidrol , Pengecatan lepehne , Leukemia One laboratory examination to ensure the presence of leukemia is conducted by cytochemical staining, including to the use of lepehne reagent which appears green on the old erythrocyte granules, used as its comparison, in coloring the aligned erythrocyte cells (eritroblas). This research aims at examining the influence of perhidrol (H2O2) concentration variations on the quality of lepehne staining. This study is classified into a quantitative true experimental research type which samples are the bone marrow biopsy results collected with a random sampling method. Based on the microscopic observation results on five samples, it is found that perhidrol concentration 30% as the control has the color of a clear concentrated green. Meanwhile, the perhidrol concentration 27.5% (v/v) and 25% (v/v) have the similar result, that is, having a good lepehne staining quality with the erythrocyte color of non-concentrated green. It shows that the concentration decreases by 27.5% (v/v) and 25% (v/v) do not influence the quality of staining results. In addition, the concentration variations of 22.5% and 20% also have the similar results, in which the quality is proven poor with the erythrocyte color of unclear green. Thus, the perhidrol concentration decreases influence more on the quality of lepehne staining. Based on the data analysis conducted using Chi-Square, the p-value is 0.000 < α (0.05) that it can be concluded that perhidrol concentration variations influence the quality of lephene staining results.
