Abstract

Penelitian ini bertujuan untuk mengidentifikasi keberadaan gen enterotoksin A S.aureus (SEA) pada susu sapi murni dengan metode Polymerase chain reaction (PCR). Penelitian ini merupakan studi deskriptif. Sampel penelitian adalah susu sapi perah yang berasal dari Kampung Susu Sapi Perah Kelurahan Gedawang Semarang. Metode penelitian meliputi isolasi bakteri S.aureus pada susu sapi murni, isolasi DNA bakteri S.aureus dan deteksi gen Staphylococcal Enterotoksin A (SEA) dengan metode PCR menggunakan primer spesifik. Sebanyak 15 sampel susu sapi murni yang digunakan dalam penelitian ini berhasil diidentifikasi sebanyak 3 isolat positif S.aureus. Hasil deteksi gen Staphylococcal Enterotoksin A (SEA) terhadap 3 isolat S.aureus menunjukkan hasil negatif keberadaan gen SEA dengan ditandai tidak munculnya pita DNA dengan ukuran gen 521 bp. Kata kunci : Gen Enterotoksin A (SEA), Susu Sapi Murni, Staphylococcus aureus The aims of this study is to identified the presented of S.aureus enterotoxin A (SEA) on the pure cows milk that conducted using Polymerase chain reaction (PCR) method. The research sample was milk dairy cattle from pure cow milk of Gedawang village, Semarang. The research method were covers S.aureus bacteria on the pure cows milk isolation, S.aureus bacteria DNA isolation and Staphylococcal Enterotoxin A (SEA) gene detection by PCR method using a specific primer. A total of 15 samples of pure cow milk used in this study were identified as 3 positive isolates of S.aureus. The results of Staphylococcal Enterotoxin A (SEA) gene detection on 3 isolates of S.aureus showed negative results of SEA genes with marked absence of DNA bands with gene 521 bp size.
