Abstract

ABSTRACT The aim of this study was to determine the inhibitory of methanol extract of Heartleaf Maderavine Madevine (Anredera cordifolia (Ten.) Steenis) to the growth of S.aureus with concentration of 75%, 80%, 85%, 90% and 95%. This was an experimental research with well diffusion method. Heartleaf Maderavine Madevine contains alkaloids, flavonoids, saponins, oleanolic acids and essential oil that can act as antibacterial. The leaf used in this study was green Heartleaf Maderavine Madevine. The leaf was washed and cut into small pieces. After that, it was dried at the temperature of 250C – 270C for five days. Once it was dried, the leaf was blended until smooth to turn it into powder and sieved with a 100 mesh size. It was soxhlet-extracted using ether. The extraction was done until it became a paste or 100% concentration then freeze-dried with a concentration per well of 100µl. The antibacterial test was performed on MHA (Muller Hinton Agar) medium and incubated at the temperature of 370C for 24 hours. The result showed that Heartleaf Maderavine Madevine leaves could inhibit the growth of Staphylococcus aureus at concentration of 75%, 80%, 85%, 90% and 95% by 13.5 mm, 15.75 mm, 17.25 mm, 19.25 mm and 21 mm. Keywords: Inhibitory Action, methanol solvent extract, S. aureus, Heartleaf Maderavine Madevine Penelitian ini bertujuan untuk mengetahui daya hambat ekstrak methanol daun binahong (Anredera cordifolia (Ten.) Steenis) terhadap pertumbuhan S.aureus pada konsentrasi 75%, 80%, 85%, 90%, dan 95%. Jenis penelitian ini adalah penelitian eksperimen dengan metode sumuran. Daun binahong mengandung alkaloid, flavonoid, saponin, asam oleanolik, dan minyak atsiri yang dapat berfungsi sebagai antibakteri, daun yang digunakan dalam penelitian ini adalah daun binahong yang hijau, dicuci bersih dan dipotong kecil – kecil kemudian dikering anginkan pada suhu 250C – 270C selama 5 hari. Setelah kering daun binahong diblender hingga halus agar menjadi serbuk, diayak dengan ukuran 100 mesh, kemudian disoxhletasi menggunakan eter, ekstraksi dilakukan sampai menjadi pasta atau dikatakan konsentrasi 100% kemudian di freeze driyer dengan konsentrasi per sumuran 100µl. Uji antibakteri pada media MHA (Muller Hinton Agar) dan diinkubasi pada suhu 370C selama 24 jam. Hasil penelitian menunjukkan bahwa daun binahong dapat menghambat pertumbuhan Staphylococcus aureus pada konsentrasi 75%, 80%, 85%, 90%, dan 95% sebesar 13,5 mm, 15,75 mm, 17,25 mm, 19,25 mm dan 21 mm.
