Abstract

Pemeriksaan aPTT berfungsi untuk menggambarkan adanya defisiensi factor pembekuan atau adanya inhibitor dalam jalur intrinsik. Serangkaian tes dari sistem koagulasi intrinsik, dimana fase kontak dari rangkaian jalur koagulasi diaktivasi sebelum direkalsifikasi dengan adanya pengganti trombosit, menggunakan plasma sitrat sebagai sampel. Sentrifugasi adalah proses pemisahan sel dengan plasma yang menjadi proses pra analitik yang mempengaruhi hasil pemeriksaan aPTT, pengaruh dari sentrifugasi yang tidak sesuai adalah meningkatnya jumlah trombosit, sehingga faktor trombosit (Pf3) yang terdapat pada trombosit akan mempercepat pembentukan fibrin. Penelitian ini bertujuan untuk mengetahui pengaruh waktu sentrifugasi terhadap nilai aPTT. Jenis penelitian ini adalah eksperimen. Sampel diambil secara acak pada mahasiswa Analis Kesehatan, dilakukan pemeriksaan aPTT sebanyak 8 kali pengulangan, plasma sitrat diperoleh dari campuran antara darah dengan Na Sitrat 3,2 % dengan perbandingan 9 bagian Na Sitrat dan 1 bagian darah, kemudian di sentrifugasi dengan waktu 5, 10 dan 20 menit sebagai perlakuan sampel. Hasil penelitian diperoleh rerata pemeriksaan nilai aPTT pada sampel yang disentrifugasi 5 menit adalah 32,76 detik, sampel yang disentrifugasi 10 menit diperoleh 34,54 detik, sedangkan pada waktu sentrifugasi 20 menit adalah 37,01 detik. Data yang diperoleh kemudian dianalisa dengan menggunakan uji one way anova. Data nilai signifikan yang didapatkan 0,224 > 0,05 sehingga dapat disimpulkan tidak ada pengaruh yang signifikan waktu sentrifugasi terhadap nilai aPTT. Kata Kunci: Waktu Sentrifugasi, Nilai aPTT aPTT examination serves to describe the presence of clotting factor deficiency or the presence of inhibitors in the intrinsic pathway. A series of tests of the intrinsic coagulation system, in which the contact phase of the coagulation line chain is activated before it is recalcified by a platelet replacement, using plasma citrate as the sample. Centrifugation is the process of separation of cells by plasma into a pre analytical process that affects the results of aPTT examination, the effect of inappropriate centrifugation is the increase in platelet count, so that platelet factor (Pf3) present in platelets will accelerate the formation of fibrin. This study aims to determine the effect of centrifugation time on aPTT values. This research type is experiment. The sample was taken randomly on the Health Analyst students, the aPTT examination was 8 times repetition, plasma citrate was obtained from a mixture of blood with Na Sitrat 3.2% with ratio 9 parts Na Sitrat and 1 part of blood, then centrifugation with time 5, 10 and 20 minutes as sample treatment. The result of the research showed that the aPTT value examination on the samples that was centrifuged 5 minutes was 32.76 seconds, the sample was centrifuged 10 minutes was 34.54 seconds, while the time of centrifugation was 20 minutes 37.01 seconds. The data obtained were then analyzed by using one way anova test. Data of significant value obtained 0,224> 0,05 so it can be concluded there is no significant effect of centrifugation time to aPTT value. Keywords: Centrifugation Time, aPTT Value
