Abstract

Examination with EDTA blood should be done immediately, if forced to delay should consider the storage time limit for each examination. One of the factors that can influence the examination of platelet count is the delay of examination more than 1 hour cause the decrease of platelet count. Cells are actively still doing metabolism, the metabolism results in the accumulation of lactate and decrease in ph. The decrease in ph causes platelets to release granular content in the form of ADP so that platelet function decreases. This study aims to determine the effect of delaying examination of blood specimens at room temperature, air conditioning room and refrigerator on platelet count. The type of research is experiment, place of research at Puskesmas Tanjung in July 2017. This research was conducted on 4 samples of whole blood of UPTD Puskesmas Tanjung employees. EDTA blood is immediately checked for the first 3 tube stored at room temperature both the AC room and the third refrigerator, every 1 hour, 2 hours and 3 hours checked again. The results of the examination of the platelet count average value on the immediate examination 271 000 cells / mmᵌ, delay 1 hour room temperature 258.500 cells / mmᵌ, 266,000 cells / mm³ AC room and 268,750 sel / mm³ refrigerator. 2 hours delay at room temperature of 250,000 cells / mmᵌ, 258,500 cell / mm³ AC room and 264,500 cell / mm³ refrigerator. 3 hours delay at room temperature 238,250 cells / mmᵌ, 253,500 cell / mm³ AC room and 259,750 sel / mm refrigerator. The lowest platelet decrease in the 1 hour refrigerator delay of 0.8% and the highest decrease in a 3 hour delay at room temperature of 12%.Statistical analysis using two way anova test obtained P > 0.05, meaning there was no effect of delay of examination of blood specimens stored at room temperature, air conditioning room and refrigerator to platelet count.
