Abstract
Human Epidermal Growth Factor Receptor 2 (HER2) is a receptor on the cell surface that has a similar structure to the Epidermal Growth Factor Receptor (EGFR) which is located on chromosome 17q21 which is encoding the transmembrane glycoprotein through tyrosine kinase activity. Over expression of HER2 in cancer cases could be seen using immunohistochemical techniques (IHC). Incubation is a period of time that is needed by antibodies to make specific bonds with the antigen. The aim of this study was to find out the expression of HER2 on immunohistochemical with concentration 1:50, 1:100 and 1:150. This experimental research was using cross sectional approach. HER2-postive breast cancer tissue study sampel with +3 stages from one organ and the same patient. HER2 staining using Strep (Avidin) Biotin Complex technique. Conclusion the result showed that the most efficient 1: 50 incubation concentration will give the best result in 30 minute incubation dye, the most efficient 1: 100 incubation concentration will give the best result in 60 minute incubation dye, the most efficient 1: 150 incubation concentration will give the best result in 90 minute incubation dye. Known that the higher the concentration, the shorter the incubation time will be and the lower the concentration requires long incubation time. Keywords: HER2, IHC, Antibody primer
