Abstract

Peripheral blood smear is one of hematology examination to identify the blood cell and to find the blood parasites. Giemsa is a common stain that is used for this test. The staining time is based on Giemsa concentration. Dilution making through estimation causes comparison between Giemsa stock and solvents are inaccurate as well as the staining time. The purpose of research is to analyze the relation of Giemsa stain and dilution towards peripheral blood smear result. This research is analytical experimental which is randomly taken from 10 people. The smears from these people are drowned by Giemsa for 10 minutes with 1:4; 1:9 and 1:19 dilution. Peripheral blood smear results from 10 smears with 1:4 dilutions show excellent results, 1:9 dilutions show 2 good smears and 8 lesser results, 1:19 dilutions are completely bad. These are because the higher Giemsa concentration, the faster chemical reaction will be occurred between Giemsa pigments and blood cells. The lower dilution, the faster staining time are needed. Based on Chi Square test shows sig 0,000 is smaller than 0,05 so as the conclusion there is a significant relation of Giemsa stain and dilution towards peripheral blood smear result.
