Abstract
Histotechnique is a method of making histologic dish of specimen through series of process until ready to be observed. The one of these processes is fixation. Fixation is the process of preserving the tissues for the same conditions as alive. The solutions used for fixation include alcohol 70% and NBF 10%. Alcohol 70% is easy to obtain, cheap and fast penetration but can pursue the tissue. NBF 10% does not damage the tissue, but the power penetration is slow. The purpose of this study to know the microscopic view of cardiac quality that was fixed with alcohol 70% and NBF 10% on Hematoxylin-Eosin staining by looking at structure, absorption and the similarites in the of preparation. The type of descriptive research by cross sectional design using 60 samples fixed using 2 solutions that are alcohol 70% and NBF 10% respectively 30 samples. Good processing shows microscopic image that do not happen to changes in the structure of cardiac muscular tissue, blue color in the nucleus of the cell is clear, red color (eosin) in cytoplasm is good, and the color of preparations is similiar. The result of the microscopic view heart preparation fixed with alcohol 70% are 100% not so good while NBF 10% showed 100% good result, so NBF 10% is better to used for the heart tissue fixation.
