Abstract
Ascaris lumbricoides egg has three layers of wall one of the albumin layer. In general, water-soluble albumin and coagulated in the heat, soluble in an aqueous salt solution, and sediment in hight concentration saline solution (salting out). Protein damage can be influenced by heat, heavy metal ions, and pH (acidity) that can cause denaturation proteins. This study was conducted to determine the effect of pH concentration and contact time of the morphology of Ascaris lumbricoides eggs. This type of research is an experimental research. Sample examination was taken from Ascaris lumbricoides infected positive faeses, then sample was given six treatments with repetition four times. Furthermore, the sample is examined under a microscope by direct methods. To know the effect of pH concentration and contact time to Ascaris lumbricoides egg morphology with Chi-Square test. The result showed that the morphology of Ascaris lumbricoides eggs were damaged at a concentration of acid pH (pH 3) with a contact time of 30 minutes, whereas the acid pH (pH 3), neutral pH (pH 7), and alkaline pH (pH 10) with a contact time of 15 minutes Ascaris lumbricoides eggs looks intact and retain their morphology as well as on neutral pH (pH 7) and alkaline pH (pH 10) with a contact time of 30 minutes. Chi-square statistical test showed a significance value of 0,004 with a significance level of 0,05 is 0,004 < 0,05 so that it can be concluded that there is the influence of pH and contact time simultaneously the morphology of Ascaris lumbricoides eggs.
