Abstract
ABSTRAK Clearing adalah suatu proses dalam pewarnaan hemtoxylin eosin yang menggunakan metode perendaman, pencelupan dan penggoyangan untuk kualitas sediaan jaringan hati yang memberikan efek terhadap kejernihan dan transparan pada saat proses identifikasi. Tujuan Untuk mengetahui kualitas sediaan jaringan hati pada proses clearing menggunakan beberapa metode (perendaman, pencelupan dan penggoyangan). Gambaran mikroskopis sediaan jaringan hati proses clearing pewarnaan hemtoxylin eosin menggunakan metode perendaman di biarkan selama 10 menit, pencelupan diangkat dalam waktu 3 detik dilakukan selama 10 menit dan penggoyangan digoyangkan dengan bentuk angka 8 selama 10 menit. Hasil pengecatan hemtoxylin eosin pada proses clearing menggunakan metode perendaman diperoleh dari 10 preparat (100%) kategori baik (3), metode pencelupan diperoleh hasil dari 9 preparat (90%) kategori baik (3) dan 1 preparat (10%) kategori kurang baik (2), dan metode penggoyangan diperoleh hasil dari 8 preparat (80%) kategori baik (3) dan 2 preparat (20%) kategori kurang baik (2). Hasil uji statistik menggunakan uji kruskal-wallis diperoleh nilai p-value <0,05 yang menunjukan bahwa ada perbedaan gambaran mikroskopis sediaan jaringan hati normal hewan coba pada proses clearing dengan menggunakan metode perendaman, pencelupan dan penggoyangan pada sediaan jaringan. Hasil yang memiliki gambaran mikroskopis sediaan jaringan yang baik adalah metode perendaman dikarenakan sitoplasma dan inti sel terlihat jelas. kata kunci : Proses Clearing, Hematoxylin Eosin, Sediaan Jaringan Hati DIFFERENCES IN QUALITY STOCKS IN THE PROCESS OF TISSUE HEART CLEARING STAINING HEMATOXYLIN EOSIN (HE) USING SOME METHOD (CROCHETERS MAN, DYEING AND ROCKING ) Hasmini 1 , Toeti Rahajoe 2 , Arya Iswara 2 1. Objective To determine the quality of liver tissue preparation in the clearing process using several methods ( soaking , dipping and rocking ) . Quality stocks tissue careful process of clearing the coloring hemtoxylin eosin using methods marinade late on leave for 10 minutes , dyeing was appointed in time 3 seconds to do for 10 minutes and rocking shaken by form number 8 for 10 minutes . Results pengec a tan hemtoxylin eosin in the process of clearing using a method of soaking obtained from 10 preparations (100%) the category of either (3) , a method of dyeing obtained results of 9 preparations (90%) categories good (3) and 1 preparations (10%) categories less good (2) , and methods rocking obtained results from 8 preparations (80%) categories good (3) and 2 preparations (20%) categories of less well (2) .
