Abstract

Pemeriksaan kadar total protein serum menggunakan alat kimia analyser. Stabilitas reagen biuret selama 1 tahun pada suhu 2 - 25° C, 6 bulan pada suhu ruang ( 25 - 35° C ) atau sebelum masa kadaluwarsa yang tertera pada label etiket. Reagen Biuret jika sudah diletakkan pada alat kimia analyser, setelah ± 2 hari ( ± 48 jam ) digunakan untuk running pemantapan mutu internal, hasil pemeriksaan keluar dari nilai rentang target. Tujuan Penelitian untuk mengetahui perbedaan kadar total protein dalam serum yang menggunakan reagen biuret yang diletakkan dalam kimia analyser segera, 24 jam, 48 dan 72 jam. Jenis penelitian adalah penelitian analitik kuantitatif. Sampel diambil secara random berjumlah 10 sampel, kemudian diperiksa menggunakan reagen yang diletakkan dalam alat kimia analyser selama segera, 24 jam, 48 jam dan 72 jam. Hasil pemeriksaan menunjukan rerata hasil kadar protein serum menggunakan reagen yang diletakkan pada alat kimia analyser segera: 6.8 gr/dl, selama 24 jam : 6.6 gr/dl, selama 48 jam : 6.3 gr/dl dan selama 72 jam : 5.9 gr/dl. Hal ini menunjukkan kecenderungan menurun antara kadar total protein dalam serum menggunakan reagen biuret yang diletakkan dalam alat kimia analyser segera, selama 24 jam, 48 jam dan 72 jam. Uji statistik Kruskal Wallis menunjukkan nilai kemaknaan 0.000 dengan taraf kemaknaan 0.05 yaitu 0.000 < 0.005 sehingga dapat disimpulkan bahwa ada perbedaan yang bermakna antara kadar total protein yang diperiksa menggunakan reagen biuret yang diletakkan pada alat kimia analyser selama segera, 24 jam, 48 jam dan 72 jam. The objective of the study was to investigate the difference in total serum protein levels using biochemical reagents placed in chemical analyzers immediately, 24 hours, 48 and 72 hours. The type of research is quantity analytical research. Samples were collected by random samples of 10 samples, then examined using reagents placed in the chemical analyzer for immediate, 24 hours, 48 hours and 72 hours. The results showed that serum protein concentrations were performed with reagents placed on the chemical analyzer immediately: 6.8 g / dl for 24 hours: 6.6 g / dl, for 48 hours: 6.3 g / dl and for 72 hours: 5.9 g / dl. This shows a declining trend between total serum protein levels using biuretic reagents placed in the chemical analyzer immediately, for 24 hours, 48 hours and 72 hours. Kruskal Wallis statistic test showed the significance value of 0.000 with significance level of 0.05 0.000 < 0.005 so it can be concluded that there is a significant difference between the total protein content examined using biuret reagent placed on chemical analyzer immediate, for 24 hours, 48 hours, and 72 hours.
