Abstract
Hematocrit examination is an examination used to measure the volume of erythrocytes. Determination of hematocrit value can be done by macro and micro. The micro-way is performed using a capillary pipette that is principled with anticoagulant blood in the centrifuge for a period of time, so that the blood and plasma cells are separated. The time and speed of a centrifuge affect the value of the hematocrit, because it can result in a low height of false or cause the blood in the capillary tube to be tilted or empty. The blood volume in the capillary tube is too small, the less dense closure of putty and the less skilled manpower also affect the hematocrit value. The purpose of this study is to find out whether there are differences in hematocrit values with different time and speed of centrifuge. The type of research used is the analytical research conducted at the Hematology Laboratory of Muhammadiyah University of Semarang. The sample was taken randomly by 9 samples from student of DIII Health Analyst of Special Program of Muhammadiyah University of Semarang class of 2015. Hematocrit examination was done with time and speed 16000 rpm 5 minutes as control, 13000 rpm 6 minutes, 10000 rpm 7 minutes and 7000 rpm 8 minutes. Based on the results of hematocrit examination with different time and velocity obtained p value 0,033 (<0,05), show that there is difference of hematocrit level which checked with different speed and time of centrifuge. The difference of these results can be seen, so it should be attempted so that each examination is done with the time and speed according to the provisions for the erythrocytes to solidify optimally.
