Abstract
Pre-analytic factors are the biggest cause of laboratory error, one of which is the use of anticoagulants. Prothrombin Time (PT) examination used a 3.2% sodium citrate anticoagulation with a ratio of 9 blood volume and 1 anticoagulant volume. The purpose of this study was to determine the effect of blood volume and anticoagulation ratio of 3.2% sodium citrate to the results of examination of Protrombin Time. This research is an experimental research. The study was conducted at RSUD Kajen. Sampling using Sampling Samples used 8 respondents, each sample was treated by varying the sample volume and sodium citrate ratio by 3.2%, and then checking Protrombin Time. The result of examination with second unit obtained the mean value of Protrombin Time respectively from the ratio of sample volume and sodium citrate 3.2% 9: 1 = 12,300, 8: 1 ratio = 12,662, 7: 1 ratio = 13,075, 6: 1 ratio = 14,000 and at a ratio of 5: 1 = 15.962. The results of One Way Anova statistical test showed a significant difference of mean value of Protrombin Time with variation of sample volume ratio and 3.2% sodium citrate. (p value = 0,000, α = 0, 05). The result of Post Hoc test shows that the difference of Protrombin Time value from 9: 1 ratio to 8: 1 p value = 0,533, ratio 9: 1 to 7: 1 p value = 0,187, ratio 9: 1 to 6: 1 p value = 0,006 and the ratio of 9: 1 to 5: 1 p value = 0,000, α = 0, 05, so it can be concluded there is no significant difference between 9: 1 ratio with 8: 1 and 7: 1, while the ratio between 9: 1 and 6: 1 and 5: 1 there is a significant difference.
