Abstract
Tuberculosis is the leading cause of death among bacterial infections in the world caused by the bacterium Mycobacterium tuberculosis. The discovery of this disease is still based on microscopic diagnosis, namely by staining with ziehl neelsen staining, namely 1% carbol fuchsin, 3% acidic alcohol, 0.1% methylene blue so that it looks red with blue background. The purpose of the study was to determine the difference in washing time of 3% acid alcohol to the results of staining on smear preparations with inundation time for 3 minutes and 5 minutes. This type of research is experimental. Samples were taken with sputum criteria for positive smear patients of two or three as many as 40 preparations. The results showed washing for 3 minutes obtained 90% bad results (18 preparations), while washing for 5 minutes obtained 95% good results (19 preparations). The Whitney Man statistical test shows a significance value of 0,000 so that the significance value is 5 0.05 so it can be concluded that there is a difference in washing 3% acid alcohol for 3 minutes and 5 minutes.
