Abstract

Prothrombin Time (PT) is an examination that use to filter hemostasis in extrinsic and joint pathways, and also can use to monitor oral anticoagulation. The examination material of PT is Plasma Citrate obtained from a mixture of blood and Na Citrate 3.8% (9: 1), which centrifuged for 15 minutes at a speed 2000-2500 g. Plasma citrate should not be stored in cold temperatures because it can activate factor VII, but in the reality of the field found that examination of PT using plasma citrate with cold temperatures. The purpose of this research is to analyze the difference between PT examination result of fresh plasma and plasma stored in 2-8°C temperature for 2-8 hours. The type of this research is analytic research. The population in this study were all patients who did the PT examination at the Lab of Clinic Prodia Semarang during January 2017, and a sample of 43 patients were taken randomly. Statistical analysis using different Wilcoxon test to analyzee the difference of variation in PT examination result between fresh plasma and plasma stored in 2-8°C temperature for 2-8 hours. Result of this research from 43 samples of fresh plasma showed that the lowest of PT 11,70; the highestwas 39.40; and the averagewas 17,1930; and standarddeviation was 6,00148 while in plasma stored in 2-8°C temperature obtained result of lowest PT was 10,00; the highestwas 37.30; the averagewas 16,3814; and the standarddeviation was 6.05283. Wilcoxon test results showed that there is a significant difference between PT examination of fresh plasma and plasma storedin 2-8°C temperature for 2-8 hours with significant value is 0.000. Plasma citrate storage at cold temperatures gives significant differences to Prothrombin Time results. Keywords : PT, fresh plasma, plasma storedin 2-8°C temperature for 2-8 hours
