Abstract

Introduction: Dental health problems are one of the most common diseases, one of which is dental caries, which can impact a person's quality of life. The main cariogenic microorganisms involved in caries is from Streptococcus genus, particularly Streptococcus mutans. There is a lot molecular methods for identifying pathogenic bacteria, such as Polymerase Chain Reaction (PCR). PCR is a technique for DNA synthesis and amplification. This method can multiply target DNA millions of times, producing large numbers of DNA copies that can be used for further analysis. Purpose: This study aims to determine the results of Streptococcus mutans DNA fragments sequence from PCR amplification dental caries from patients at RSGM, Muhammadiyah University of Semarang. Method: The descriptive laboratory study, analyzing the DNA sequence of the Streptococcus mutans glucosyltransferase gene in dental caries patients at Muhammadiyah University of Semarang Hospital. The primary data from bacterial identification, Streptococcus mutans gtf gene profile, secondary data from patient medical records. Result: Based on the sequence analysis of DNA fragments from PCR amplification, a DNA band measuring ±585 bp was detected in samples 1 and 2. The size of the DNA band corresponds to the characteristics of the gtf-B gene in Streptococcus mutans. Conclusion: This study concluded that both S. mutans samples analyzed had the gtf-B glucosyltransferase gene type.
