Abstract

ABSTRAK Pemeriksaan hitung jumlah trombosit dilakukan dengan metode manual cara langsung dengan metode tabung menggunakan bilik hitung pada 12,5 kotak sedang. Jumlah trombosit dalam keadaan normal sangat dipengaruhi oleh cara menghitungnya mengingat sifat-sifat trombosit cenderung melekat, mudah menggumpal, sukar dibedakan dengan kotoran kecil. Antikoagulan EDTA mengikat ion kalsium dengan menghambat pembentuskan thrombin untuk mengkonfersi fibrinogen menjadi fibrin, sama halnya dengan ekstrak batang mangrove yang memiliki senyawa flavonoid yang bersifat anti pembekuan darah mengikat ion kalsium sehingga proses transfer Ca2+ ke dalam sitoplasma sel platelet dihambat oleh senawa flavonoid dan turunan lainnya sehingga tidak terjadi agregrasi platelet. Penelitian ini bertujuan untuk mengetahui perbedaan hasil jumlah trombosit menggunakan antikoagulan EDTA dan ekstrak batang Mangrove. Penelitian ini dilakukan terhadap 16 sampel darah dengan antikoagulan EDTA dan antikoagulan ekstrak batang mangrove mahasiswa DIV Analis Kesehatan lintas jalur kelas D. Setiap sampel dilakukan pemeriksan hitung jumlah trombosit. Pengujian data dalam penelitian menggunakan uji Shapiro-Wilk. Hasil rata-rata antikoagulan EDTA 307000/mm³ dan hasil rata-rata antikoagulan ekstrak batang mangrove 258000/mm³. Dilanjutkan uji Independent Sampe T-Test dengan nilai (sig(2-tailed). 0.090) menunjukkan tidak ada perbedaan yang bermakna antara jumlah trombosit menggunakan antikoagulan EDTA dan ekstrak batang mangrove (Aegiceras corniculatum). EDTA anticoagulants bind calcium ions by inhibiting thrombin formation to convert fibrinogen to fibrin, as well as mangrove extract extracts that have anti-clotting blood-binding compounds that bind calcium ions so that the process of transfer of Ca2 + into the cytoplasm of platelet cells is inhibited by the flavonoid senawa and other derivatives so No platelet aggregation occurred. This study aims to determine differences in the results of platelet counts using EDTA anticoagulants and Mangrove stem extracts. This study was conducted on 16 blood samples with anticoagulant EDTA and anticoagulant mangrove stem extracts of DIV Health Analyst students across Class D pathways. Each sample was tested for platelet count. Testing data in research using the Shapiro-Wilk test. The average anticoagulant yield of EDTA is 307000 / mm³ and the average anticoagulant yield of mangrove stem extract is 258000 / mm³.
